Weekly enrichment (2026-07-20)
- The open-label extension (MIT News, 14 Nov 2025; published in Alzheimer’s & Dementia) followed 5 volunteers who continued daily 40 Hz light-and-sound GENUS stimulation for about two years after MIT’s early-stage clinical trial, the longest-term test of the noninvasive method to date.1
- The 3 late-onset AD participants (all female) scored significantly higher than age/sex/cognition-matched controls drawn from national databases on 3 cognitive tests, and showed increased brain-wave responsiveness and improved circadian-rhythm measures at 30 months.1
- In the 2 late-onset volunteers who provided plasma, phosphorylated tau (pTau217) fell 47% and 19.4% on a test recently approved by the FDA as the first plasma biomarker for diagnosing Alzheimer’s.1
- The 2 early-onset male participants showed no significant cognitive benefit and significantly reduced brain-wave responsiveness, leading the authors to attribute the differential response to disease onset rather than gender.1
- The original 2020 MIT trial enrolled 15 mild-AD volunteers for one hour/day of 40 Hz stimulation delivered via an in-home LED panel and speaker; though cut short by Covid-19, it found significant benefits at three months (preserved brain volume was not significant at the 30-month point).1
- The approach is being scaled by MIT spinoff Cognito Therapeutics, whose pivotal Phase 3 HOPE trial (NCT05637801) is the largest AD-focused medical-device pivotal trial, enrolling ~670 mild-to-moderate AD patients at 70 U.S. sites for 60 min/day of Spectris AD stimulation over 12 months, with primary completion estimated June 2026.2 3
- Cognito’s earlier OVERTURE study reported a 76% reduction in cognitive decline (MMSE), a 77% reduction in functional decline (ADCS-ADL), and roughly a 69% reduction in whole-brain atrophy over 6 months.3
- GENUS (gamma entrainment using sensory stimuli) is designed to entrain 40 Hz gamma to target amyloid, tau, and neuroinflammation; it holds FDA Phase 3 status for Alzheimer’s and is also being explored in Parkinson’s disease and Down syndrome.4
